You cannot drive directly from Singapore to Malaysia by land alone. The Johor Strait separates the two; crossing requires the Johor–Singapore Causeway or the Malaysia–Singapore Second Link (bridge), or a ferry/train. Both bridges carry road traffic, so you can drive across them, but there is no continuous land connection without crossing the strait.
